This kit is based on the property of DNA to adsorb to silica surfaces in the presence of high concentrations of chaotropic salts. The lysis of cells from swabs is performed in the lysis solution containing chaotropic salts and nonionic detergents. The lysis process is supported by auxiliary, high affinity protease (Proteinase K). Such reagents assure lysis and complete protein degradation. Subsequently the Iysate is loaded onto the spin minicolumn with dedicated silica membrane. The DNA binds to the silica membrane, while contaminants (fragmented proteins, salts. etc.) pass through the column freely. Residual contaminants are moved away by a wash step. Pure DNA is eluted with a low salt buffer or water and can be used in following experiments without further purification procedures (e.g. precipitation).
