日本荣研麻疹、腺病毒酶联免疫诊断试剂盒
检验原理
用抗原包被微量板孔,制成固相载体。加患者血清到板孔中,其所含的抗体特异性地与固相载体中现存抗原结合,形成免疫复合物。除去多余物质后,加入结合了碱性磷酸酶的IgG、IgA或IgM抗体,使之与上述免疫复合物反应。洗板,除去多余的结合物,加入底物(对硝基苯磷酸盐)。其与酶结合的免疫复合物反应,产生有颜色产物,颜色强度与特异性抗体含量成正比。

霍乱弧菌能还原硝酸盐为亚硝酸盐,靛基质反应阳性,当培养在
含硝酸盐及细菌氨酸的培养基中,产生靛基质与亚硝酸盐,在浓
硫酸存在时,生成红细菌,称为霍乱红反应,但其他非致病性弧
菌亦有此反应,故不能凭此鉴定霍乱弧菌。EL Tor型霍乱弧菌与
古典型霍乱弧菌生化反应有所不同。前者Vp阳性而后者为阴性。
前者能产生强烈的溶血素,溶解羊红细胞,在血平板上生长的菌
落周围出现明显的透明溶血环,古典型霍乱弧菌则不溶解羊红细
胞。个别EL Tor型霍乱弧菌株亦不溶血。
抗原性
根据弧菌O抗原不同,分成Ⅵ个血清群,第Ⅰ群包括霍乱弧菌的两
个生物型。第Ⅰ群A、B、C三种抗原成份可将霍乱弧菌分为三个血
清型:含AC者为原型(又称稻叶型),含AB者为异型(又称小川
型),A、B、C均有者称中间型(彦岛型)。
抵抗力
霍乱弧菌古典生物型对外环境抵抗力较弱,EL-Tor生物型抵抗力
较强,在河水、井水、海水中可存活1~3周,在鲜鱼,贝壳类食
物上存活1~2周。
霍乱弧菌对热,干燥,日光,化学消毒剂和酸均很敏感,耐低温
,耐碱。湿热55℃,15分钟,100℃,1~2分钟,水中加0.5ppm
氯15分钟可被杀死。0.1%高锰酸钾浸泡蔬菜、水果可达到消毒目
的。在正常胃酸中仅生存4分钟。能发酵葡萄糖、麦芽糖、甘露醇
、蔗糖、半乳糖等产酸产气,迟缓发酵乳糖。不分解阿拉伯糖、
水杨素、卫矛醇、鼠李糖、木糖、侧金盏花醇和肌醇。氧化酶、
明胶液化试验阳性。V-P试验大多数EL-Tor生物型阳性,而古典
生物型大多为阴性。霍乱弧菌能分解细菌氨酸产生吲哚,同时能
还原硝酸盐为亚硝酸盐。因此当霍乱弧菌培养在含硝酸盐的蛋白
胨水中,所产生的亚硝酸盐与吲哚结合成亚硝基吲哚,滴加浓硫
酸即出现蔷薇细菌,称为霍乱红试验阳性。
