纳米金Gold Nanoparticles的制备方法主要采用还原剂,如柠檬酸钠,硼氢化钠等还原氯金酸HAuCl4·4H2O氯金酸HAuCl4·4H2O在还原剂的作用下,可以聚一定大小的金纳米颗粒,形成带负电的疏水胶溶液。由于静电作用而成为稳定的胶体状态,故称胶体金Colloidal gold

纳米金随粒径的变化而颜色发生变化:不同尺寸形貌的纳米金对可见光有选择性吸收,吸收了某个波长的光后,将没有吸收的光反射出去,我们看到的就是反射的光,其实不是他们的颜色,不过是他们将这种颜色的光反射给我们看到了,这种光在我们的视觉中是某种颜色而已。不同尺寸形状的颗粒,它们的阻尼现象不同,一般来说小颗粒的阻尼现象,从而小颗粒振动频率要小于大颗粒,它们也就吸收长波的光,反射短波的光。随着颗粒增加,它们吸收的光波波长变短,也就是所谓的红移现象了。

1 纳米金应用于毛细管电泳检测尿液中8-OHdG

8-OHdGDNA氧化的主要物,体液中8-OHdG为终产物,稳定性,因此尿中的8-OHdG成为评价个体变危险或与自由基相关的生物标记物,

2纳米金应用于蛋白质纤维染色的研究

   纳米金溶胶应用于对羊绒、羊毛纤维及羊毛织物进行染色,探讨了纳米金与蛋白质的作用原理。(早孕试纸:HCG胶体金法检测试剂盒采用双抗体夹心一步法技术,以胶体金为指示标记,检测尿液中的HCG浓度)

3 纳米金应用于的研究进展

   金纳米粒子具有的生物亲和性及的表面性质,经表面修饰的金纳米粒子通过共价或非共价的方式与靶向分子或者相连,获得多功能纳米粒子,应用于的靶向与。金纳米粒子是一种的靶向制剂,可以同时实现被动靶向和主动靶向:有纳米粒径大小引起的通透性和滞留效应(enhanced permeability and retention effect , EPR )可以实现被动靶向,使纳米金载体在组织选择性分布,以增加并减少副作用。此外,金纳米粒子进行表面修饰后,可利用靶向分子实现的主动靶向,将或射线特异带到部位,靶向的目的。纳米金团簇表面的边缘位点和顶部位点4f轨道的结合能相对较低,这些位点形成的Au-S键的键能也相对较低,容易发生离解,这对Au-MMPCs的稳定性和配体交换有影响.Au-MPCs具有粒径小和生物相容性好的,用巯基或二硫化合物对Au-MPCs表面进行修饰,形成具有生物活性的Au-MMPCs,这些生物活性Au-MMPCs同待转运物和膜相互作用,通过静电吸附和内吞作用可将转运物传送和核内,靶向的传输和释放需要依靠Au-MPCs的作用.

4 纳米金生物探针及其应用

    纳米金颗粒的表面不表现为静电作用,还有疏水作用。为了让蛋白质与纳米金颗粒结合,减小静电作用,而是疏水作用起作用。通过表面改性,让纳米金带正电,从而与带负电的蛋白质相结合,制成用于电子显微镜的金探针。胶体金与抗人球蛋白(anti human-IgC)结合制备的金探针可以定性检测艾滋(HIV)抗体。在纳米金微粒上修饰DNA探针,由于共价结合力,在DNA探针的一段连接上巯基后可与纳米金结合,通过识别颜色的变化,就能检测出相应的靶DNA

5纳米金在生物标记分析中的应用进展

   金纳米粒子除具有一般纳米粒子的表面效应,量子尺寸效应外,同时具有的生物相容性。由于生物大分子和功能化同处于纳米维度当在相应的生物体系中加入功能化的纳米金颗粒使其具有定向吸附效应以及富集效应等。两者通过化学或物理吸附可制得既稳定又具有一定计量关系的生物/纳米复合材料。该复合材料又可以用来构建具有功能的分子结构体系,此外也可以作为检测探针用于研究复杂的生物化学体系。

6 纳米金在生物传感器中的应用

   胶体金增DNA生物传感器的检测灵敏度,纳米级的金颗粒标记在DNA分子上,可增加该分析分子的质量,从而增质量性。胶体金能提供一似于能保持蛋白质稳定性的,实习附在其表面的蛋白质保持他们的生物活性,能增乳酸生物传感器的检测灵敏度。用金纳米粒子修饰铂电而制成的葡萄糖氧化(GOD)生物传感器,具有灵敏度,速度快等。
