将外源基因导入真核细胞是分子生物学研究中的一项重要技术。转基因载体主要分为病毒载体系统和非病毒载体系统。病毒载体系统转染效率高,是体内基因治疗的主要工具,但安全性存在隐患,且有免疫原性,体内不能反复应用,制备较复杂,而且所能装载的外源DNA大小有限,使其应用受到很大限制。非病毒载体是病毒载体的重要补充途径,一般认为安全性高,载体一般无免疫原性,而且是基于质粒的转染,易于组装[1-2]。目前已有的非病毒型载体介导基因转染的方法有裸DNA注射、磷酸钙介导、电转移法、阳离子脂质体及阳离子聚合物介导等。1995年,Boussif*报道了聚乙烯亚胺(polyethylenimine,PEI)可作为非病毒载体,其后,聚乙烯亚胺成为阳离子多聚物研究zui快的领域。为了研究PEI基因转染试剂的转染效果,本文通过PEI转染表达绿色荧光蛋白的pEGFP质粒到293T细胞,以观察其效果,并测定了多种因素对转染效率的影响。
