The SciFlow-1000 system is composed of two fluidic divisions; see Figure 1.
? Wells 1 and 2 are the fluid reservoir and fluid regulator; these wells
work in tandem. The Source well supplies the row with fresh media.
As the media enters the system, the regulator well helps maintain
consistent fluid levels across the plate.
? Wells 3 – 11 are locations for cells.
? In the first generation SciFlow1000, well 12 is configured to
incorporate additional functionalities as the product line grows. This
protocol is written so that fluid should not enter this well 12.
SciFlow is designed to combine three main features for creating biological
systems on the benchtop:
1. Constant fluid renewal
2. Compartmentalized well-to-well communication
3. Gradient-over-time exposures
Users can take advantage of any or all of these features depending on the
specific biological questions
To use the SciFlow plate, users can follow typical cell culture
approaches to plating cells and changing media while maintaining isolated
culture wells. Once it is desired to link wells together, creating a system of
connected cultures users simply increase the volume of media in culture
