单细胞全基因组测序是在单细胞水平上对全基因组进行扩增与测序的一项技术，是近年来研究最为热门最为前沿的技术之一。原理非常简单，首先将要研究的单个细胞分离出来进行全基因组DNA扩增，获得高覆盖率和完整的基因组之后进行高通量测序，进而揭示单个细胞间的基因差异。因此，在扩增过程中获得高覆盖率以及高保真性的扩增产物是得到全面准确测序结果的必要条件。
传统的PCR方法在扩增过程中容易出现较多的扩增偏倚、基因缺失等现象，尤其是在单细胞扩增过程中容易丢失大量序列。而近年来发展出来的MDA（Multiple Displacement Amplification,多重置换扩增）技术不仅具有高分辨率和高基因组覆盖度，而且具有更好的敏感性和特异性，因而是gong认较好的单细胞基因组扩增技术。下面就为大家介绍一款来自Expedeon公司基于MDA技术的单细胞全基因组扩增试剂盒TruePrime® Single Cell WGA Kit。
TruePrime®采用MDA技术，基于新型的TthPrimPol 引物酶和Phi29 DNA聚合酶，扩增过程无需添加随机引物，减少了扩增偏倚，提高了扩增过程的覆盖率和保真性，并且对于污染的外源DNA不敏感，可重复性好。TruePrime®可用于单个细胞或少量细胞的全基因组扩增，与常见的NGS平台如Illumina和IonTorrent等兼容性良好。
