QuantiTect Probe RT-PCR Kit 提供含有 HotStarTaq DNA Polymerase、ROX参比染料和 dNTP混合液的预混液。为便于使用，预混液可储存在2-8°C条件下。dNTP混合液含有dUTP，可进行UNG处理。该试剂盒同时提供经优化的RT混合液（储存在-20°C条件下），可高效、灵敏地对RNA模板进行逆转录，模板量适用范围广。

原理
Real-time RT-PCR过程中产生的非特异性产物和引物二聚体会与特异性产物竞争反应组分，从而干扰目标基因的准确定量。QuantiTect Probe RT-PCR Kit克服了这些问题，通过使用*的RT-PCR缓冲液促进引物、探针和PCR模板之间高特异性退火 （见图"Specific annealing"）。此外，缓冲液中包含HotStarTaq DNA 聚合酶，相对其他聚合酶，该酶能提供更严谨的热启动（见图"Highly specific amplification"）。

操作流程
QuantiTect Probe RT-PCR Kit无需进行繁琐耗时的反应条件优化。只需简单地将引物、探针和模板加入到即用型PCR预混液中，即可在各种定量PCR仪上开始反应 （见流程图 和表"Components of the QuantiTect Probe RT-PCR Kit"）。

应用
QuantiTect Probe RT-PCR Kit包括HotStarTaq DNA聚合酶和*的RT-PCR 缓冲液，能够高灵敏度定量低拷贝的RNA靶序列，并且可实现宽线性范围的精确定量（见图 "High sensitivity and efficiency, and wide dynamic range"）。在同一个管中连续进行逆转录和PCR不会影响灵敏度，通过与两步法定量 RT-PCR的 CT 值对比可证明（见图 "One-step RT-PCR with comparable performance to two-step RT-PCR"）。

One-step RT-PCR with comparable performance to two-step RT-PCR. Tenfold serial dilutions of cDNA or RNA （100 ng to 1 pg） prepared from leukocytes were analyzed in duplicate on the Mx3005 using primers and a FAM-labeled probe specific for IL1R2 （interleukin 1 receptor, type II）.  A  Two-step RT-PCR using the QuantiTect Probe PCR Kit （PCR efficiency: 101%）.  B  One-step RT-PCR using the QuantiTect Probe RT-PCR Kit （PCR efficiency: 104%）.  C  The CT values from the 2 amplification plots.
