APOEε4等位基因是zui常见的阿兹海默症遗传风险基因。不过空气污染物与阿兹海默症之间的关系的却很少有人注意。而墨西哥城是一个快速发展伴随严重环境污染的城市,800万城市生活的儿童都不得不每天暴露在PM2.5超标的环境中。为了评估APOEε4健康儿童在认知,嗅觉和代谢性脑指数方面是否受到空气污染物影响,研究人员对50个生活在墨西哥城市的13.4±4.8岁携带APOEε3或者APOEε4的孩子进行了韦氏儿童智力量表(WISC-R)评分和气味鉴别实验,同时用质子磁共振测量海马,脑桥,额叶顶叶白质的脑代谢产物。如预测一样,携带APOEε4的孩子右额叶白质的关键代谢产物比例比APOEε3的儿童要少,对肥皂的臭味鉴别困难,言语和智商平分低于平均分,并且显现出短期记忆和对关注的显著递减。

近日,来自英国的科学家在生物学期刊molecular and Cellular biology在线发表了他们关于RNA聚合酶III调控巨噬细胞功能的研究进展。
研究人员指出,肿瘤微环境中的炎症具有许多促癌症发展效应。特别是肿瘤相关巨噬细胞(TAM)能够产生许多细胞因子,促进恶性肿瘤细胞存活,血管生成以及转移维持肿瘤生长。肿瘤相关巨噬细胞内细胞因子的合成与蛋白质合成过程紧密相关,而蛋白质的翻译过程又依赖于RNA聚合酶III转录形成的众多tRNA。因此研究RNA聚合酶III对肿瘤相关巨噬细胞的功能发挥有何影响具有重要意义。

The APOEε4 allele is the most common genetic risk factor for Alzheimer's disease. However, little attention has been paid to the relationship between air pollutants and Alzheimer's disease. Mexico City is a rapidly developing city with severe environmental pollution, and 8 million children living in cities have to be exposed to excessive levels of PM2.5 every day. To assess whether APOEε4 healthy children were affected by air pollutants in terms of cognition, olfactory and metabolic brain indices, the researchers performed Wechsler’s inligence on 50 children with APOEε3 or APOEε4 at ages of 13.4±4.8 years living in Mexican cities. Table (WISC-R) scores and odor identification experiments were performed. Proton magnetic resonance was used to measure brain metabolites in the white matter of the hippocampus, pons, and frontal lobe. As predicted, the proportion of key metabolites in the right frontal white matter of children with APOEε4 is lower than that of APOEε3 children. It is difficult to identify soap odors, scores of speech and IQ are lower than average, and short-term memory and attention are shown. Significantly decreasing.
The researchers believe that the continuous exposure to urban air pollution caused poor perception, changes in the metabolism of glial cells and neurons. Living in a heavily polluted city, the combined effects of APOEε4 may lead to an acceleration of neurodegenerative changes. Air pollution is a serious public health problem. Exposure to airborne pollutant concentrations that meet or exceed current standards has been shown to be related to neuroinflammation and neuropathology. In the United States, 200 million people live in environments with excessive ozone and fine particulate matter. Moreover, there has been evidence that particulate pollutants are significantly associated with increased mortality from stroke, cardiovascular and respiratory diseases. There are more health problems related to children living in major cities in Mexico.
There is an urgent need for a more in-depth study to prove that the interaction between APOE ε4 and air pollution can affect children's brains, and may provide new and unprecedented tips for the prevention of Alzheimer's disease in the future.
Recently, scientists from the United Kingdom published their latest research on RNA polymerase III regulating macrophage function in the online journal of molecular and cellular biology.
The researchers pointed out that inflammation in the tumor microenvironment has many cancer-promoting effects. In particular, tumor-associated macrophages (TAMs) are capable of producing many cytokines, promoting the survival of malignant tumor cells, and angiogenesis and metastasis to maintain tumor growth. The synthesis of cytokines in tumor-associated macrophages is closely related to the process of protein synthesis, and the process of protein translation depends on the numerous tRNAs that RNA polymerase III transcribes. Therefore, it is of great significance to study the effect of RNA polymerase III on the function of tumor-associated macrophages.
