线粒体是人类细胞将糖转化成能量的重要工厂。西奈山伊坎医学院的科学家们开发了一种新测序技术，可以帮助人们理解线粒体基因对个体患病风险的影响，比如糖尿病、心脏病和癌症。这项研究发表在近期的Nucleic Acids Research杂志上。
在年龄、体重和坏习惯(比如吸烟)相同的人中，有些会生病有些却不会。研究者们一直对这一点感到迷惑，并尝试通过饮食和核基因来确定个体的患病风险。但这样的分析遗漏了线粒体基因(mtDNA)的差异，mtDNA遗传自我们的母亲，每个细胞都有这样的DNA。

Mseek主要是利用酶清除细胞核DNA，只留下线粒体DNA进行测序。研究人员将Mseek用于多个细胞系，鉴定了不同细胞样本中的mtDNA“指纹”。他们还通过追踪mtDNA变体发现，mtDNA异质性在单细胞水平上是稳定维持的。

Mitochondria are important plants where human cells convert sugar into energy. Scientists at the Icahn School of Medicine in Mount Sinai have developed a new sequencing technology that can help people understand the impact of mitochondrial genes on individual risk factors such as diabetes, heart disease and cancer. This study was published in the recent issue of Nucleic Acids Research.
Some people who are the same in age, weight, and bad habits (such as smoking) are sick but not others. Researchers have always been puzzled by this and tried to determine the risk of the individual through diet and nuclear genes. But such an analysis missed the difference in mitochondrial genes (mtDNA) that we inherited from our mother, and each cell has such DNA.
Recent studies have shown that mistakes in mitochondria can cause serious diseases. However, due to the heterogeneity of mtDNA, it is very complicated to analyze the impact of mtDNA mutation on individual disease risk.
This study shows the first sequencing technology that can accuray identify mtDNA heterogeneity, Mseek. The mtDNA content in the cells is very small, and there has been little progress in this area. People are aware of the heterogeneity of mtDNA but can no longer establish deterministic theories at the single cell level.
“Investigators are always looking for low-cost methods that can accuray sequence mtDNA,” said Dr. Ravi Sachidanandam, an assistant professor at Icahn Medical School in Mount Sinai. "Now we have done this. Mseek can identify functional disorders in mitochondria, making mtDNA a useful biological indicator. In addition, people can use mtDNA as a potential target for the treatment of cancer and many genetic diseases."
According to Dr. Sachidanandam, the new sequencing technology has achieved unprecedented sensitivity, specificity, and sequencing depth, enabling high-purity mtDNA to detect different versions of mtDNA or mtDNA variants.
Mseek uses enzymes to remove nuclear DNA, leaving only mitochondrial DNA for sequencing. The researchers used Mseek for multiple cell lines and identified mtDNA "fingerprints" in different cell samples. They also found that by tracking mtDNA variants, mtDNA heterogeneity was stable at the single cell level.
